Para passar da negativa para a afirmativa tira-se o not depois do verbo (AM/ IS/ ARE)
Ex. The girl is not happy = negativa
      The girl is happy = afirmativa
Para passar da afirmativa para a negativa é só fazer o contrário (acrescentar o not depois do verbo)

Para vc passar da afirmativa para a interrogativa tem que colocar o verbo no inicio da frase e acrescentar o ponto de interrogação:
Ex. The boy is sad = afirmativa
      Is the boy sad? = interrogativa
